ICT’s assay diluents are proprietary buffer formulations designed to equalize any differences between the sample matrix (serum, plasma, urine, cell culture fluid) and the diluent used to generate the standard curve. These diluents also reduce nonspecific interactions between the sample matrix proteins and the plate surface which translates into lower background noise.
Assay diluents are pipetted directly onto the plate just prior to adding the samples. They may be used with neat or diluted samples, depending on the characteristics of the sample matrix and the target range of the assay. Since different sample matrices have unique characteristics when run in an ELISA, they must be matched with specific assay diluents. Use of an assay diluent reduces the effects of the sample matrix and variation among samples, without pre-dilution of the samples.

ICT's General Assay Diluent reduces the matrix differential between the serum/plasma sample and the diluent used to generate the standard curve. AD1 contains proteins from goat sera to reduce nonspecific interactions that might occur between the sample matrix proteins and the plate surface. It also contains a proprietary calcium chelating agent designed to inhibit interference from complement and thrombin present in serum and plasma samples.

ICT’s IgM Reducing Assay Diluent is formulated for use with problematic serum and plasma samples tested in antibody sandwich ELISA formats. Proprietary additives have been included in AD2 to reduce IgM-mediated conjugate bridging interference which can otherwise cause substantial background noise. AD2 contains proteins from goat sera to reduce nonspecific interactions that might occur between the sample matrix proteins and the plate surface. It also contains a proprietary calcium chelating agent designed to inhibit interference from complement and thrombin present in serum and plasma samples.
Antigen Down and Sandwich ELISAs; for serum and plasma samples tested in all antigen-down ELISAs; for human, porcine, and bovine serum samples.
ICT’s Neptune Assay Diluent contains a non-mammalian protein base designed to minimize nonspecific binding of the sample matrix to "sticky" adsorbed protein antigens. The formulation contains specially selected detergents to enhance the specific signal without denaturing immobilized proteins. This is especially useful when evaluating human, porcine, and bovine serum samples. AD3 also contains proprietary additives designed to inhibit complement and thrombin activity present in serum and plasma samples. Neptune Assay Diluent does not contain any immunoglobulin-based components that could interfere with anti-IgG signal-generating formats.
Antigen Down and Sandwich ELISAs; for serum and plasma samples tested in all antigen-down ELISAs; to eliminate clotting of plasma samples in the ELISA plate well.
ICT's Antigen-Down Assay Diluent is specifically formulated for testing plasma and serum samples in antigen-down ELISA formats. Proprietary additives, including EDTA, have been incorporated into this diluent that eliminate the risk of plasma sample clots in the ELISA plate well while also minimizing nonspecific binding interactions associated with these complex matrices.
